Advanced medicines have revolutionised the treatment of many diseases. Our current allocation at Arrhenius, transferred over from Dardel, has been used for large scale data analysis to improve the safety and production of advanced medicines. Now, we would like to combine our current allocation with previous projects on Cryo-EM from Berzelius which was based on discovering new advanced medicines. Using the Arrhenius resource at NSC, we can perform research to improve the efficacy, safety and production of proteins and viruses for medicine use.
The Arrhenius resource will be used for primarily two purposes: (1) to analyse RNA, DNA and Proteomic data; (2) analyse structures in Cryo-EM.
The RNA, DNA and Proteomic data arise from several different types of projects. Some projects are involved in improving the production of advanced medicines from mammalian production cells in a bioprocessing setting. By analysing RNA, DNA and proteins from different cell lines with different efficacies in medicines production, we can determine factors which could be modulated to increase production. We also have projects analysing the DNA from viruses produces for advanced medicine purposes, to better understand if only the desired DNA is encapsidated and not unwanted DNA.
Via previous allocations at Berzelius, we have also made significant progress in finalising structures of viruses and proteins intended for use in advanced medicine. However, due to the new admission rules at Berzelius, we were unfortunately unable to finish the projects to the point of publication. We therefore ask for resources at the Arrhenius cluster to finalise the structural analysis.
We want to precisely determine the molecular interactions between a newly developed protein and its cellular target, a cell receptor clinically relevant for cancer. We also want to structurally determine a part of a flexible region of a virus for use as advanced medicine in gene therapy. We are certain we can accurately resolve these two issues using the Arrhenius resource. We will use an installed version of RELION on the cluster or CryoSPARC via PReSTO once this resource is available.
