Abstract

The purpose of this project is to identify genetic variants and environmental factors that influence risk for developing psychiatric disorders. Rather than studying a singular disorder, our collaborative research group at the Karolinska Institute is taking a multidisorder approach. We are simultaneously studying several major disorders and outcomes, including obsessive-compulsive disorder, Tourette syndrome, major depressive disorder, eating disorders and suicide death in work supported by multiple Swedish funding bodies. Our study utilizes two major Swedish resources: the national PKU Biobank, which houses newborn blood samples from nearly all individuals born in Sweden since 1975, and Psychiatry Sweden, a register created by linkage of several national and local registers in Sweden, via the unique personal identification number. To date, we have extracted DNA from a total of ~10,000 newborn blood spots for the conditions listed above, as well as healthy controls. The DNA is currently being microarray genotyped, and we request a Bianca project space to perform quality control of these data, genotype imputation and genome-wide assocation study (GWAS). Our group has several years of experience performing GWAS on Bianca using established pipelines. We expect to genotype several thousand more samples in the coming year and are seeking funding to add other data types, such as DNA methylation and exome sequencing.
