Abstract

We intend to use the tool AlphaPulldown for an "all against all" folding of predicted proteins to learn about protein-protein interactions during a Mycobacterial phage infection. This dataset will be used to correlate with size exclusion DIA-MS datasets which our lab has generated during various time points of phage infection cycles. This work aims to validate the protein-protein interactions important throughout the phage infection and to better understand the numerous poorly characterized proteins important for successful phage, with hope of translating this into clinically relevant therapeutic strategies. Our initial proof of the concept will be done with phage D29 and its host Mycobacteria smegmatis, for which we already produced SEC-DIA-MS data. We have several other phage-host pairs in the pipeline which we will test under varying conditions in the future.
