This project investigates transcriptional changes in healthy and diseased rat and mouse kidneys to understand the metabolic and mitochondrial control of interstitial and epithelial cells in healthy and diseased kidneys on a single cell level using single-cell RNA sequencing technology. We use a pharmacological approach to manipulate the hypoxia-inducible factor transcription factor pathway/mitochondrial axis, which regulates cellular metabolism and mitochondrial function in kidney cells. Single-cell RNA sequencing will be performed by the SNP&SEQ Technology Platform, National Genomics Infrastructure (NGI), Science for Life Laboratory at Uppsala University. Spatial transcriptomic sequencing will be performed by the Royal Institute of Technology (KTH), NGI, SciLifeLab in Stockholm. The project will provide information on oxygen-regulated metabolic and mitochondrial responses in individual kidney cell populations under healthy and disease conditions.
