Glioblastoma is the most aggressive of all brain tumours, with a 5-year survival rate as low as low as 1.2%. Recent research emphasises that neuronal synapses, especially glutamergic synapses impact positively the proliferation of glioblastoma. Through this project we would like to dissect specific genomic and transcriptomic alterations in glioblastoma when co-cultured with different neuronal types. To understand the impact and the complexity of the neuronal inputs, the co-culture was treated with picrotoxins drug that affects neuronal activity and SIS3, SMAD3 transcription factor inhibitor, or both. Glioblastoma from the co-cultured cells were taken extracted and samples under different conditions were processed for RNA-Seq or ATAC-Seq library preparation. The analysis of the data will reveal changes in gene expression and open chromatin landscape in glioblastoma influenced by different neuronal inputs.
