Abstract

Existing techniques for studying the epigenome are constrained by limitations in acquiring comprehensive genomic information with spatial precision. In recent years, novel platforms have been developed to address this issue. In line with these advancements, we have recently introduced a pioneering method called spatial ATAC, leveraging 10X Genomics' OMNI platform, which enables us to conduct spatially resolved epigenomic analysis. Our recent research demonstrates the efficacy of this technique in elucidating the regulatory mechanisms governing spatial gene expression patterns during mouse development. However, it is worth noting that the aforementioned platform is currently inaccessible to the general research community and lacks multi-omics capabilities. Consequently, we aim to address these two limitations by adapting the same technique to the widely available transcriptomics Visium platform. Our objective is to achieve comparable resolution to the published datasets and ensure its applicability to both young and adult tissues. By doing so, we can broaden the accessibility and utility of this spatial epigenomics approach.
