This project investigates transcriptional changes in healthy and diseased rat kidneys to understand the metabolic and mitochondrial control of interstitial and epithelial cells in healthy and diseased kidneys on a single cell level using single-cell RNA sequencing technology. We use a pharmacological approach to manipulate the hypoxia-inducible factor transcription factor pathway/mitochondrial axis, which regulates cellular metabolism and mitochondrial function in kidney cells. Single-cell RNA sequencing will be performed by the SNP&SEQ Technology Platform, National Genomics Infrastructure (NGI), Science for Life Laboratory at Uppsala University. The project will provide information on oxygen-regulated metabolic and mitochondrial responses in individual kidney cell populations under healthy and disease conditions.
