Abstract

Single-cell RNA sequencing (scRNA-seq) makes it possible to study the hematopoietic system based on the gene expression levels of single cells. Such high-resolution analyses have shed doubt on the classic hierarchical models of hematopoiesis. Moreover, while recent studies have led to an improved picture of the cell differentiation landscape, some differentiation trajectories—such as that of the mast cell—remain poorly understood. By borrowing tools and concepts from optimal transport (OT) and systems theory, and applying them on single cell multiomics data, this project seeks to further our understanding of the cell differentiation landscape. In particular, the aim is to chart likely differentiation trajectories from hematopoietic stem cells to lineage-committed progenitors, and to infer cell lineage-forming potential of individual multi- or bipotent progenitors.
