Abstract

Glioblastoma GBM) is a grade IV brain tumor that is fatal with median survial of 15 months. It is molecularly heterogeneous, highly aggressive and has high heterogeneity not only between patients, but even within the same patient. This makes diseaae modelling in in-vitro challenging. The project will establish a human GBM (glioblastoma)-brain organoid model pipeline, in which multiple patient derived GBM cell lines ( from the HGCC cell culture repository; hgcc.se ) are grown on / with brain organoids of different age. The growth, establishment and invasion pattern of the GBM cells will be tracked by microscopy, providing tumor growth parameter phenotyping. Simultaneously, at designated time points, GBM-organoid samples will be collected, isolated and screened by scRNAseq to provide molecular phenotypes and state for the different. Mutation analysis and methylation analysis will be performed and overlayed on the scRNA and imaging data once the initial scRNA analysis is in place.This pipeline will help form a backbone for personalised patient organoid culture and drug screening.
