Abstract

Arabidopsis thaliana mutants with altered genes encoding core spliceosome proteins exhibit symptoms that are significantly enhanced by suboptimal temperatures. We postulate that temperature and other environmental conditions may have an impact on plant's splicing capacity. We will measure the splicing capability in wild-type and other mutant genotypes at various temperatures to test this theory. We will combine metabolic RNA labelling and high-throughput strand-specific RNA sequencing to ascertain the splicing capability of plants. In addition, we will examine the impact of temperature on splicing and transcript usage using long-read single-molecule RNA sequencing of nascent transcripts. Thus, this project at Uppmax will be used to conduct the data analysis on this project.
