Abstract

An epidemiological study has also shown that first-born girls have a greater risk of developing overweight and obesity compared to their younger sister, a second-born girl. A hypothesis for this is believed to be maternal constraint which mainly affects the first child. Identification of changes in gene expression in the placenta that indicate that the child has been exposed to fetal programming leading to an increased risk of later morbidity, could mean an opportunity for early prevention and treatment. Gene expression in placenta from two consecutive pregnancies will be analyzed with biological co-variates including sensitive data will be analyzed. In collaboration with NBIS who will be responsible for • transfer of the raw reads data to Bianca (the user will transfer the count and normalized tables to Bianca as well as the metadata (BMI, age etc. of the pregnant women at the time of each pregnancy)) • Quality control of the data • Paired differential expression analysis without adjustment for covariates (including multiple testing correction) • Discussion with the group about initial results and about which covariates should be included as adjustment variables in further adjusted paired differential expression analyzes • Gene set/pathway enrichment analysis • Delivery: report including detailed method description and visualization of the results as well as list of differentially expressed genes from each analysis. The code used for the analysis will also be provided.
