Abstract

In this project we will map the biodistribution of different variants of DNA origami nanostructures in zebrafish. We have previously mapped the biodistribution of fluorescently labelled DNA origami nanostructures in zebrafish embryos using light-sheet microscopy. We found that the nanostructures injected in day-old embryos accumulate at the caudal hematopoietic tissue (equivalent to the embryonic liver) within minutes unless coated with PEG-polylysine, which enables the widespread detection of nanostructures within zebrafish larvae up to 3 days post-injection. In this project we will perform single cell sequencing of zebrafish embryos treated with fluorescently labelled DNA origami nanostructures to analyse their biodistribution in detail.
